There's a recurring myth in oral solid dosage development: one disintegrant is enough. In theory, yes. In practice — especially for high-dose, poorly soluble, or hydrophobic APIs — a single disintegrant often underdelivers on disintegration time, particularly under accelerated stability conditions.
The smarter strategy is a synergistic disintegrant system: Sodium Starch Glycolate (SSG) as the primary superdisintegrant, paired with a controlled level of Sodium Carboxymethyl Cellulose (CMC-Na) to modulate water uptake and wicking.
How the synergy works
- SSG swells rapidly on contact with water, generating the mechanical force that breaks the tablet matrix.
- CMC-Na controls the rate and direction of water penetration, preventing the "gelling lock" that sometimes traps the API inside a swollen shell.
- The result: faster, more reproducible disintegration — even at lower total disintegrant load.
This combination is particularly effective in:
- Orally disintegrating tablets (ODTs)
