Two of the hardest problems in solid oral dosage have nothing to do with the API and everything to do with how powder behaves in your equipment and how the drug releases in the patient.

Problem one: poor powder flow. Fine APIs, sticky blends, and high-dose formulations don't flow through your hopper or fill your die uniformly. The fix is usually fumed silica (colloidal silicon dioxide) at 0.1–0.5% — a tiny addition that changes everything.

Problem two: release profile control. Whether you need sustained release, delayed release, or once-daily dosing, the workhorse polymer remains Hydroxypropyl Methylcellulose (HPMC) — a hydrophilic matrix former with decades of regulatory acceptance.

Used together, they form a quietly powerful pair: fumed silica ensures your blend fills consistently, while HPMC ensures the resulting tablet releases API exactly as your dissolution spec demands.

  • Fumed silica acts as a glidant and anti-caking agent, reducing angle of repose and improving die-fill uniformity — critical for high-speed presses.
  • HPMC (particularly the 2208 and 2910 substitution types) forms a robust gel layer that controls drug diffusion, allowing you to tune release from 2 hours to 24 hours.
